How to Reconstitute Peptides: Step-by-Step Guide

What Is Reconstitution?

Lyophilised (freeze-dried) peptides must be dissolved in a sterile liquid before use. This process is called reconstitution. The most commonly used diluent in research settings is bacteriostatic water (BW) — sterile water containing 0.9% benzyl alcohol to prevent microbial growth.

What You Need

Step-by-Step Process

  1. Calculate your desired concentration using the CitySupps Reconstitution Calculator.
  2. Wipe the rubber stopper of both vials with an alcohol swab and allow to air dry.
  3. Draw the calculated volume of bacteriostatic water into your syringe.
  4. Inject the BW slowly into the peptide vial, directing the liquid down the side of the glass — never directly onto the powder.
  5. Gently swirl (do not shake) until the powder is fully dissolved. The solution should be clear.
  6. Label the vial with the date of reconstitution.

Storage After Reconstitution

  • Store at 2–8°C (standard refrigerator)
  • Keep away from light
  • Use within 28 days
  • Do not freeze the reconstituted solution
